STAT1 Drives Senescence in the Aging Retina

Integrated transcriptomic and proteomic profiling of aged mouse retinas identified STAT1 as a convergent regulatory hub linking oxidative stress, inflammatory signaling, and cellular senescence. In human retinal pigment epithelial cells, forced STAT1 expression was sufficient to induce senescence-like programs and reactive oxygen species, while STAT1 knockdown blunted oxidant-driven senescence and downstream p53 and inflammasome activation. The retina offers an accessible readout of how inflammatory aging programs initiate in specific cell populations and spread across tissue.

Key Points

  • STAT1 emerged as central hub coordinating retinal aging programs
  • STAT1 overexpression alone triggered senescence and reactive oxygen species
  • Knockdown reduced oxidant-induced senescence, p53, and inflammasome signaling

Longevity Analysis

Oxidative load and low-grade inflammatory signaling converge on a single transcriptional node in neural tissue, which means reducing cumulative oxidative burden has consequences beyond local antioxidant status — it shapes whether senescence programs are initiated at all. Expression localized to horizontal cells and glia while target programs appeared across multiple cell types, illustrating how damage in one compartment propagates inflammatory cues through an entire tissue. For visual function, which degrades predictably with age and is rarely regained, the actionable layer sits upstream: limiting oxidative exposure and supporting the cellular machinery that clears damaged components before senescent states consolidate.
