Senescent Cells Spread Aortic Valve Calcification Cell to Cell

In calcific aortic valve disease, senescent valvular interstitial cells hypersecrete mitochondrial-derived vesicles that transmit a pro-calcific signal to neighboring cells, allowing calcification to spread from isolated foci through the valve. The senescence-associated secreted factor Gas6 drives the final scission step of vesicle release via a calcium–CaMKII–DRP1 axis, and blocking either the kinase, the fission protein, or Gas6 itself reduced calcification, stenosis, and fibrotic remodeling.

Key Points

  • Senescent valve cells release mitochondrial vesicles that calcify neighboring cells
  • Gas6 drives vesicle scission through calcium–CaMKII–DRP1 signaling
  • Gas6 deletion reduced valvular stenosis, calcification, and fibrosis in vivo

Longevity Analysis

Aortic valve calcification has long been treated as passive mineral deposition, but this work frames it as an actively transmitted cellular state — damaged mitochondria packaged into vesicles that carry osteogenic instructions from one cell to the next. The link between cellular senescence, mitochondrial quality control, and structural stiffening of the heart's outflow tract places valve disease squarely within the same biology that governs vascular aging and tissue regeneration capacity. Targeting the secreted signals of senescent cells, rather than the calcium deposits they produce, offers a mechanistic entry point upstream of surgical valve replacement.
