Optogenetic Retinal Therapy Holds Vision Gains at Four Years

Nanoscope Therapeutics will present phase 2 randomized controlled data on sonpiretigene isteparvovec optogenetic therapy for geographic atrophy and four-year extension data on MCO-010 in retinitis pigmentosa at the Euretina Congress in Vienna. The intravitreal optogenetic approach is mutation-agnostic, requires no genetic testing or surgery, and is delivered as a single dose. A biologics license application for MOGENRY has been accepted and filed with the FDA following RESTORE phase 2b/3 results.

Key Points

  • Four-year extension data show sustained vision improvement with MCO-010
  • First randomized phase 2 optogenetic data in macular degeneration
  • Single intravitreal dose, no genetic testing or surgery required

Longevity Analysis

Vision loss is among the most functionally consequential age-related declines, restricting mobility, cognitive engagement, and social participation long before it threatens survival. Optogenetic therapy repurposes surviving retinal cells to carry light signals the degenerated photoreceptors can no longer transmit, restoring a sensory channel rather than merely slowing its loss. Durability over four years from a single office-based injection is the more instructive signal here, since sustained functional restoration without repeat intervention is the standard against which regenerative approaches to age-related tissue failure should be judged.
