HPK1 Degrader Enters Clinic as Oral Checkpoint Alternative

Arvinas has dosed the first participant in a Phase 1/2 trial of ARV-6723, an oral PROTAC that degrades hematopoietic progenitor kinase 1, a brake on T-cell activation, in adults with advanced solid tumors. The study evaluates the compound alone and with pembrolizumab in patients who have already progressed on checkpoint inhibition. Preclinical work reported single-agent antitumor activity in seven models where HPK1 inhibition or anti-PD-1 therapy alone produced no benefit.

Key Points

  • First HPK1 PROTAC degrader to enter clinical testing in US
  • Targets patients who progressed after checkpoint inhibitor therapy
  • Degradation outperformed enzymatic inhibition across seven preclinical tumor models

Longevity Analysis

Removing a protein entirely rather than blocking its active site addresses a limitation that has constrained immune-directed therapy: many regulatory proteins exert effects through scaffolding functions that survive enzymatic inhibition. HPK1 restrains T-cell signaling, and degrading it restores the immune surveillance capacity that declines with age and allows malignant cells to persist undetected. Oral dosing and the focus on patients who have already exhausted checkpoint blockade point toward a more precise reading of why immune recognition fails in a given individual, rather than escalating the same intervention.
